Delaney first gained public prominence through Twitter, where he began tweeting in 2009. He had over 1.2 million followers by 2016. While other comedians were afraid to post their material on social media, he is regarded as one of the first to publish jokes on social media. He attributed his breakthrough to Irish comedic writer Graham Linehan, who began reacting to his tweets. Delaney was voted one of the ten funniest people on Twitter by Paste magazine in 2010. He was the first comedian to receive the “Funniest Person on Twitter” category at The Comedy Awards in May 2012.

Delaney shot a pilot for a variety program called @RobDelaney for Comedy Central in December 2011. The series, however, was not picked up. Instead, Delaney went on to co-write and co-star in Catastrophe, which premiered on Channel 4 in the UK on January 19, 2015. The show follows their characters after they meet after a brief affair when he is on business in the UK and then move there permanently after he finds she is pregnant. It was released on Amazon in the United States in June 2015. After four seasons, the show was canceled. On February 12, 2019, Channel 4 showed the last episode, while Amazon revealed the US release date.

Delaney appeared in a number of Hollywood blockbuster blockbusters throughout 2019. He worked again with filmmaker David Leitch and actor Dwayne Johnson on Hobbs & Shaw, playing Agent Loeb. In Paul Feig’s comedy-drama Last Christmas, he played a theatre director. Both films debuted at the top of the box office in the United Kingdom. He also played Todd opposite Anne Hathaway in The Hustle and Charlize Theron in the Fox News drama Bombshell. He appeared as Elvis Presley in Dexter Fletcher’s Elton John musical biopic Rocketman, but his appearance was deleted from the theatrical version. It was also rumored that he would co-star with Reynolds again in Pokémon: Detective Pikachu. He did not, however, feature in the finished film.
