Matarazzo started acting at the age of six; after taking over the microphone at an AIDS benefit for children, she was given the card of a talent management, with whom she worked for ten years. In 1997, she received an Independent Spirit Award for her role as adolescent social outcast Dawn Wiener in Welcome to the Dollhouse. Matarazzo has expressed appreciation for the opportunity to play interesting characters, some of whom “are ostracized for various reasons.” She has stated that her most proud performance was in 1999’s Our Guys: Outrage in Glen Ridge, which was based on a true tale and starred her as a mentally challenged girl who is raped by footballers.

She played Heather Wiseman in the short-lived television series Now and Again. She played Lilly Moscovitz in the teen romance comedy film The Princess Diaries in 2000, and she returned to the role in the 2004 sequel The Princess Diaries 2: Royal Engagement. Matarazzo has been on several popular television shows, including Roseanne, Law & Order, The L Word, Grey’s Anatomy, and Strangers With Candy. Matarazzo appeared opposite Thaao Penghlis in the world premiere of Charles Evered’s play Class at Cape May Stage in Cape May, New Jersey, in May and June 2010. Matarazzo revealed in 2011 that she would begin production on her directorial debut, a television series based on Diane Hanks’ memoir, Summer Camp: A Memoir.
