Littleford worked in improvisational theatre with the Chicago City Limits group, organized a sketch comedy team, and developed and performed her own one-woman show, This Is Where I Get Off, with the Circle Repertory Company. Littleford is arguably most known for her groundbreaking work as one of The Daily Show’s first female correspondents. Littleford’s portions on the show were satirical celebrity interviews in the vein of Barbara Walters, and utilized soft-focus filming for humorous effect, with almost nothing visible to the audience. Her appearance on The Daily Show drew the attention of actor/producer Michael J. Fox, a fan of the show. Fox starred in and executive produced the ABC sitcom Spin City.

After appearing on The Daily Show, she landed a recurring role as Deirdre West on Fox’s Spin City. She has also appeared as a guest star on several television shows since the late 1990s, including The West Wing, Family Guy, and Frasier. Littleford also appeared as a celebrity commentator on VH1 shows such as I Love the 80s Strikes Back in 2003, I Love the 90s in 2004, I Love the 90s: Part Deux in 2005, I Love the New Millennium, and The Great Debate in 2009. She was the female protagonist in the short-lived Fox series Method and Red, and she also played Ben Tennyson’s mother Sandra in Ben 10: Race Against Time, which she reprised in Ben 10: Alien Force episode 20 and Ben 10: Ultimate Alien.
