Spiridakos relocated to Vancouver in 2007 to pursue acting, and she landed her first television role, a small part on Supernatural, within weeks. She continued to work in television, appearing as a guest star on Bionic Woman, The L Word, Hellcats, and Psych. Spiridakos appeared in the television film Goblin as well as the web series Mortal Kombat: Legacy. She also appeared as werewolf Brynn McLean on Being Human. Her first starring role was as 15-year-old Becky Richards in the Canadian Teletoon series Majority Rules! in 2009. She made her feature film debut in 2011 with Rise of the Planet of the Apes, and she co-starred in the low-budget Michael Greenspan thriller Kill for Me with Donal Logue and Katie Cassidy.

She went on to play Charlotte “Charlie” Matheson, a survivalist in a dystopian future civilization, in the NBC television series Revolution. She auditioned for the role while in Los Angeles for her first pilot season. Spiridakos filmed the pilot episode in Atlanta and the first season in Wilmington, North Carolina. For her performance in the first season, she was nominated for a Saturn Award for Best Actress on Television, but she lost to Fringe’s Anna Torv. For the second season, production was relocated to Austin, Texas. Spiridakos appeared as Dawn, Morning Randolph’s daughter, in Season 3 of the Showtime sitcom Episodes. Spiridakos took part in the Ice Bucket Challenge in August 2014 to help raise awareness of the ALS disease.

Spiridakos debuted as Annika Johnson, a prostitute who arrives at the hotel at the start of the third season, on the A&E television drama Bates Motel in 2015. She will also appear in Finola Hughes’ romantic comedy Byrd & the Bees, opposite Jonathan Rhys Meyers. Spiridakos starred in an untitled CBS television pilot directed by Pamela Fryman in 2015. She plays Holly, a childhood friend who reconnects later in life, opposite Adam Brody. Spiridakos guest-starred in the final three episodes of the fourth season of the NBC police drama series Chicago P.D. as Detective Hailey Upton, a former Robbery-Homicide detective who joins the Intelligence Unit, before being promoted to series regular for the fifth season.
