Moffett starred opposite John Malkovich in a New York City stage production of Balm In Gilead after appearing in numerous Chicago stage productions. To avoid confusion with British actor Donald Moffat, he adopted the stage name “D. W.” He made his Broadway debut in The Real Thing in 1984. In 1985, he played a gay man dying of AIDS opposite Brad Davis in the original stage production of Larry Kramer’s The Normal Heart. The same year, he appeared in a Broadway production of The Boys of Winter opposite Matt Dillon. He also appeared in the television film An Early Frost, alongside Aidan Quinn and Gena Rowlands, about two male lovers during the HIV/AIDS crisis.

In Bob Rafelson’s thriller Black Widow, Moffett made his feature film debut. He later appeared in the thriller film Lisa (1990), playing a serial killer with whom a teenage girl falls in love, unaware that he is a dangerous predator. That same year, he appeared in the psychological horror film Pacific Heights as a supporting actor. He also appeared alongside Tracy Pollan in NBC’s made-for-television film adaptation of Danielle Steel’s novel Fine Things (also 1990). Moffett went on to star in Bernardo Bertolucci’s Stealing Beauty (1996), opposite Liv Tyler, Jeremy Irons, and Rachel Weisz, and in Molly, starring Elisabeth Shue, in 1999. Moffett first appeared on the NBC (and later, WB) sitcom For Your Love in 1998.

Moffett appeared in Steven Soderbergh’s crime thriller Traffic (2000) as part of an ensemble cast, earning him a Screen Actors Guild Award for Outstanding Performance by a Cast in a Motion Picture. Later, he appeared in the drama Thirteen (2003) as the absent father of a troubled adolescent girl (played by Evan Rachel Wood) and co-starred in the thriller Twisted (2004) as the attorney of a serial killer. Moffett appeared in The Philadelphia Story, opposite Kevin Spacey, at London’s Old Vic theater in 2005. He played Bob Hardy in the CW Network series Hidden Palms and Dr. Danny Clarke in the 2007 CW series Life Is Wild. From 2008 to 2010.

He appeared as a co-star in the NBC drama series Friday Night Lights. Moffett appeared as Elliot, an on/off love interest of the main character played by Fran Drescher, in the comedy series Happily Divorced from 2011 to 2012. In the ABC Family series Switched at Birth, he played John Kennish, Bay’s and Toby’s father, as well as Daphne’s biological father, from 2011 to 2017. Moffett was named chair of the Savannah College of Art and Design’s film and television program in 2017.
