Santos worked as a dresser in the costume department at the Oregon Shakespeare Festival for several years. He went to San Francisco in 2001 and spent eight years working at various luxury retail establishments in Union Square while doing stand-up comedy and frequenting open mics directly after work. Later, he came to Los Angeles to pursue a stand-up career, but he struggled to grow owing to comedy club politics. In 2012, he began appearing as a panelist on the late-night talk show Chelsea Lately while also working as a restaurant host. Santos returned to acting in 2014 when he was selected for the annual CBS Diversity Showcase.
He quickly found himself being invited to more and more auditions as a consequence of the showcase, which is given to a host of industry experts including agents and casting directors, finally securing him a position on the NBC comedy television show, Superstore. He made his Superstore debut in Fall 2015 as Mateo Liwanag, a gay, undocumented Filipino commonly misidentified as a Mexican retail store clerk. In 2017, Santos appeared at the Bridgetown Comedy Festival. He appeared as a supporting character in the 2018 romantic movie Crazy Rich Asians. In the spring of 2019, Santos appeared in the Off-Broadway production Happy Talk. Jesse Eisenberg is the author. Santos was confirmed to have been cast in Guardians of the Galaxy Vol. 3 in June 2022.
