Henderson debuted as Stuart Neilson in the New Zealand prime-time soap opera Shortland Street in 1992, at the age of 17. He remained in the role until 1995. Shortland Street became the longest-running drama and soap opera in New Zealand. He then spent three years in Australia, appearing in a number of Australian films and television shows, including Echo Point and Home and Away. Henderson immigrated to the United States in 1997 on a student visa to pursue a career in Hollywood films and to train in a two-year program at the Neighborhood Playhouse in New York City. He took a night job as a restaurant food runner to help pay his tuition. He played Hamlet in the off-Broadway play Ophelia Thinks Harder while still in high school.
Heath Ledger assisted him financially during a period of numerous auditions with no success. Henderson first appeared in a Fanta commercial as a voice actor with an Australian accent. In 1999, he starred in Kick, for which he received an AACTA Award nomination for Best Actor in a Supporting Role in 2000. After more than a year of unsuccessful auditions in Los Angeles, he was cast in a supporting role in the John Woo-directed war film Windtalkers in 2001. Henderson co-starred in the horror film The Ring with Naomi Watts in 2002. Following the success of the film at the box office, he was cast as Drew in Perfect Opposites and then as the lead character in the biker film Torque.
He was set to co-star with Jordana Brewster in Mr. & Mrs. Smith, a television series based on the 2005 film. The series, however, was not picked up by any network. In 2006, he starred in the film Flyboys. He also appeared in the House episode “Painless” and a commercial for the 2008 Cadillac CTS. Henderson was cast as a New Zealander doctor in Grey’s Anatomy creator Shonda Rhimes’ short-lived television series Off the Map in 2010. He appeared in the multi-platform Australian drama television series Secrets & Lies in 2014, as well as the SundanceTV drama series The Red Road. Henderson co-starred in Everest with Jake Gyllenhaal in 2015. In June of that year, he was cast as a potential love interest on Grey’s Anatomy.
The actor left the show two years later, with his last appearance in October 2017. Henderson told Deadline that his decision to leave was motivated by storytelling, adding, “This [was] my final year, so I was expecting Nathan’s storyline to be wrapped up.” He co-starred in the 2016 film Miracles from Heaven with Jennifer Garner. Henderson has been a co-star on the Netflix series Virgin River with Alexandra Breckenridge since 2019, playing Jack Sheridan, a former marine with PTSD. Netflix has renewed the series for a fourth and fifth season in 2021. He played Wayne Gilroy in the A24 horror film X (2022).
