Chambers was noticed on the Paris Metro by a modeling scout and went on to star in advertisements for Calvin Klein’s fragrance line. He worked for Calvin Klein, Armani, and Dolce & Gabbana in Europe, Japan, and the United States. He moved to New York City in the early 1990s after deciding on an acting career and studying at the HB Studio. For four years, he worked at Studios and Ron Stetson Studios. His first major acting role was in the Dave Matthews Band music video “Ants Marching” in 1995. He went on to have a few television roles, including one as the original Nicholas Hudson in the NBC soap opera Another World and New York Undercover.
He was cast as a series regular in the CBS primetime soap opera Four Corners in 1998. Chambers has also appeared in a number of TV movies, including Harvest of Fire, Rose Hill, and Seasons of Love. Chambers appeared in several films, including Liberty Heights (1999, as Trey, co-starring with Adrien Brody), The Wedding Planner (2001, as Massimo, co-starring with Jennifer Lopez), The Musketeer (2001), Southern Belles (2005), The Zodiac (2005), and the HBO original film Hysterical Blindness (2002). Chambers was cast as one of the leads in the CBS crime drama Cold Case in 2003, but left after only three of the first four episodes.
Chambers was cast as surgical intern Alex Karev in Shonda Rhimes’ medical drama series Grey’s Anatomy in August 2004, alongside Ellen Pompeo, Sandra Oh, Katherine Heigl, and T. R. Knight. In 2006, he and other cast members received a Screen Actors Guild Award for Outstanding Ensemble Performance in a Drama Series. He also appeared as a guest star on Grey’s Anatomy spin-off Private Practice in 2009 for a Grey’s Anatomy/Private Practice crossover event. Chambers officially announced his departure from Grey’s Anatomy on January 10, 2020, and that his final episode aired on November 14, 2019, with a farewell episode airing on March 5, 2020.
Chambers is involved with the Pentecostal Christian Church Mission Dream Center of Los Angeles, a non-profit organization. He revealed in late January 2008 that he had a sleep disorder and had checked himself into UCLA Medical Center to treat exhaustion after only two hours of sleep per week. Chambers does hot yoga.
