Wilkinson grew up in Wheaton, Illinois, and graduated from Nicolet High School in Milwaukee, Wisconsin. Wilkinson wrote for herself as a child and co-created a satirical newsletter with her pals in high school, but she never took writing seriously. She attended Harvard University and studied economics, with plans to write for television. Wilkinson joined The Harvard Lampoon writing staff as a freshman, and in her junior year, she was chosen president of the publication, becoming the first African American woman to occupy the post.
